Instrumentation

Dionex ICS 3000 Dionex ICS 3000-HPLC: Equipped with an electrochemical and conductivity detector, dual gradient pump and AS 50 autosampler. It is primarily used for the composition analysis of monosaccharides, uronic acids, sialic acids after separation by high pH anion exchange chromatography. Pulsed Amperometric Detector is capable of detecting nM amount of monosaccharides and oligosaccharides. Analysis of Sulfate and Phosphate are done using the IonPac column and conductivity detector. This instrument is also equipped with online desalter for collection of appropriate fractions for further analysis.
Dionex DX-500:Equipped with a GP-40 quaternary gradient pump, a thermostated autosampler ThermoSeparations AS3500, a Dionex RF2000 variable wavelength fluorescence detector and Shodex RI-101 detector. This system is utilized primarily for analysis of sialic acids by reverse phase chromatography following derivitization with DMB and of 2-AB tagged or untagged oligosaccharides. This system is compatible for handling both high salt and organic mixture of solvents enabling the use of reverse phase, normal phase and strong ion-exchange columns. Fraction collection is possible using this system.

Dionex DX-600 BioLC: Equipped with a GP-50 quaternary gradient pump, AS 50 autosampler, Dionex RF2000 variable wavelength fluorescence detector. This system is affixed with a post-column addition pump and a heated post-column reactor, primarily used for the analysis of glycosaminoglycan-derived disaccharides. Detection is based on post-column fluorescent derivatization using 2-cyanoacetamide.

Dionex DX-600 BioLC: Equipped with a GP-50 quaternary gradient pump, AS-50 autosampler, LC30 chromatography enclosure, AD 25 variable wavelength UV detector and an ED50 pulsed amperometric detector. This system is also capable of handling aqueous and organic solvents and is primarily used for monosaccharide, sugar nucleotide, various UV absorbing compound identification and quantitation.

HP5890Gas Chromatography-Mass Spectrometry (GC-MS): The GC-MS system is equipped with Agilent Technologies 7820A gas chromatograph system with Rtx-5ms capillary column (30m x 0.25mm x 0.25┬Ám) and coupled to 5975 mass selective detector. This instrument is very sensitive and can detect pM amount of samples and is primarily used for monosaccharide and linkage analysis of glycans. This instrument is also used for fatty acid and amino acid analysis upon request from clients.

QSTAR NLQqTOF Maldi-Mass MALDI-qTOF: Applied Biosystems QSTAR XL QqTOF with ms/ms System: The instrument is a hybrid quadrupole time-of-flight mass spectrometer, this configuration is ideally suited for the analysis of all classes of glycoconjugates including free glycans, glycopeptides, and glycolipids. This instrument is specifically used for N-linked and O-linked oligosaccharides in both positive and negative mode. The tandem mass fragmentation is done using Argon as collision gas.

 LC-MS: An LTQ Orbitrap Discovery electrospray ionization mass spectrometer (Thermo Scientific) equipped with Thermo Scientific Ultimate 3000 HPLC system. This system also has an online Ultimate 3000 variable wavelength detector. This system is primarily used for sulfated oligosaccharide analysis generated from glycosaminoglycan using a reverse-phase capillary column.
